My clinical and translational research interest is to advance our understanding of SAPHO (Synovitis, Acne, Pustulosis, Hyperostosis, and Osteitis) syndrome, Chronic Nonbacterial Osteomyelitis (CNO) and Chronic Recurrent Multifocal Osteomyelitis (CRMO), an understudied and disabling autoinflammatory disease, with no FDA-approved therapies. I am investigating disease activity, symptom patterns and skeletal involvement with advanced imaging (MRI), patient-reported measures of key quality of life and outcome domains, treatments, long-term complications, and differences between the genes in adults with SAPHO-CNO/CRMO. As the principal investigator (PI) of the adult SAPHO-CNO Study (SCS), a prospective observational study, and working closely with Dr. Polly Ferguson (world-leading expert in CNO/CRMO), we aim to address critical knowledge gaps in adults with SAPHO-CNO. The Lenert Lab is funded through the NIH and National Institute of Arthritis and Musculoskeletal and Skin Diseases K23 grant. Through our ongoing work, our goals are to improve disease activity assessments in the clinic, and to identify potentially modifiable factors and symptom groups that are predictive of skeletal damage for earlier intervention. Additionally, the discovery of mutations in inflammatory pathway genes may shed light on the spectrum of disease in adult SAPHO-CNO, and these findings could be used to guide future clinical trials testing different treatment strategies in our patients.
